Safety and efficacy data from CheckMate 568 and CheckMate 227 supported the approval of the CheckMate 9LA regimen. Progression-free survival (PFS) and overall response rate (ORR) by blinded independent central review (BICR) were also hierarchically-tested as secondary endpoints. The primary efficacy outcome measure was overall survival (OS). Patients on the control arm with non-squamous histology could receive optional maintenance therapy with pemetrexed 500 mg/m 2 IV every 3 weeks until disease progression or unacceptable toxicity. Patients with squamous histology received carboplatin IV AUC 6 plus paclitaxel IV 200 mg/m 2 every 3 weeks and patients with non-squamous histology received pemetrexed 500 mg/m 2 plus investigator’s choice of carboplatin AUC 5 or 6 or cisplatin 75 mg/m 2 every 3 weeks. The approval of nivolumab with ipilimumab in combination with platinum-doublet chemotherapy (nivo/ipi/chemo) was primarily based on CheckMate 9LA, an open-label trial in patients with metastatic or recurrent NSCLC without prior therapy for metastatic disease randomized 1:1 to nivolumab 360 mg IV every 3 weeks with ipilimumab 1 mg/kg IV every 6 weeks plus two cycles of histology-based platinum-doublet chemotherapy or four cycles of histology-based platinum-doublet chemotherapy. This review was conducted under Project Orbis, a global collaborative review program launched by FDA’s Oncology Center of Excellence (OCE) to facilitate concurrent submission, review, and regulatory action for high-impact clinically significant marketing applications across multiple participating countries ( 14). Herein, we provide a summary of FDA’s review of the marketing application that led to the approval of nivolumab, ipilimumab, and chemotherapy for first-line treatment of advanced NSCLC. Importantly, the regimen includes only two cycles of chemotherapy compared to existing combinations of immune checkpoint inhibitors with four cycles of chemotherapy. The approval of nivolumab, ipilimumab, and two cycles of platinum-doublet chemotherapy marked the first treatment regimen combining two immune checkpoint inhibitors for patients with advanced NSCLC, regardless of PD-L1 status. There are also several frontline chemotherapy sparing options for PD-L1-selected populations ( 8– 13). In patients with metastatic NSCLC without targetable genomic tumor aberrations in EGFR, ALK, ROS1, or BRAF, standard-of-care first-line treatment options include immune checkpoint inhibitors administered in combination with four cycles of histology-based platinum-doublet chemotherapy ( 4– 7). Metastatic NSCLC accounts for significant mortality globally, and effective and durable treatment options represent a significant unmet medical need. Most patients present with metastatic disease and long-term survival is poor ( 3). In the U.S., approximately 229,000 adults will be diagnosed with lung cancer in 2020, of which approximately 80–84% of cases will be NSCLC ( 3). Lung cancer remains the leading cause of cancer deaths worldwide, despite recent reports of decreased mortality with advances in treatment ( 1, 2). The benefit-risk analysis supports FDA’s approval of nivolumab with ipilimumab and chemotherapy. This was the first NSCLC application reviewed under FDA’s Project Orbis, in collaboration with Singapore’s Health Sciences Authority (HSA), Australia’s Therapeutic Goods Administration (TGA), and Health Canada (HC). Progression-free survival (PFS) and overall response rate (ORR) per blinded independent central review (BICR) were also statistically significant. Overall survival (OS) was improved for patients who received nivolumab with ipilimumab and chemotherapy, with a median OS of 14.1 months (95% CI: 13.2, 16.2) compared to 10.7 months (95% CI: 9.5, 12.5) for patients who received chemotherapy (HR 0.69 96.71% CI: 0.55, 0.87 p = 0.0006). The approval was based on results from Study CA2099LA (CheckMate 9LA), an open-label trial in which 719 patients with NSCLC were randomized to receive nivolumab with ipilimumab and two cycles of chemotherapy (n=361) or four cycles of platinum-doublet chemotherapy (n=358). Food and Drug Administration (FDA) approved nivolumab with ipilimumab and two cycles of platinum-doublet chemotherapy as first-line treatment for patients with metastatic or recurrent non-small cell lung cancer (NSCLC), with no epidermal growth factor receptor (EGFR) or anaplastic lymphoma kinase (ALK) genomic tumor aberrations.

That being said, the fact that the M1A can pump out M61s or similar rounds at a firing rate of 700 RPM makes it obvious to include in the conversation of top guns. While there is one round from each of the aforementioned calibers above M61 statistically (7.62x51 M993 & 7.62x39 7N37), neither of those are purchasable on the game’s dynamic trader market, making M61 the most viable and practical round of choice. The base ergonomics are actually lower on the M1A as well, but this bad boy has something that number 10 doesn’t: 7.62x51 NATO rounds.ħ.62x51 M61 is notably one of the most lethal rounds in the game, and the stats back that up. Similar to the SVD, the M1A is a heavy-duty marksman rifle that is most lethal on the longer end of the mid-range group, with an effective range clocking in slightly under that of the SVD. Next on the list is another one of our trusty DMRs - the M1A. Options for 20 round magazines, creating more room for scav massacreĩ.High caliber bullets that can instantly kill enemies if placed correctly.Modifications allowing for either long, mid, or even close-range use.900-meter effective range (Sniper Rifle Equivalent).Needless to say, the SVD is one long-range killer that you do not want to be on the receiving end of. The gun is even so powerful, that one of the game’s several AI Bosses, Shturman, uses it as his chosen weapon. The SVD’s more modernized design allows for it to be fully modded if necessary, granting even more options for usage, depending on your preferences. What makes the SVD stand out though is that it can shoot these same high-caliber rounds in a semi-automatic fashion. The rifle uses the same ammunition as the now-infamous Mosin, a bolt-action sniper rifle that itself does plenty of work. The SVD is a DMR (Designated Marksman Rifle)/Sniper Rifle hybrid, and packs one hell of a punch if you’re the mid/long-range type. That being said, here are 10 of the best performing weapons you can find in Escape from Tarkov. In order to evade bands of ruthless mercenaries and maybe even grab some loot in the process, you will have to be fully equipped with the best arsenal around. While it may seem easy, especially if, like myself, you’re a weathered veteran of one or both genres, escaping Tarkov with your life will often take more than just losing your dignity along the way. In Escape from Tarkov, a perfect blend between FPS and survival, you have one main objective: get out alive. And all the action and all the emotionality of the story is really built around him answering that question.” While discussing Sam Wilson in this upcoming sequel, Onah said to Collider, “We’re really testing him, and now that he’s taken the mantle of Captain America, he’s really going to question what it means to lead. New World Order will be helmed by Nigerian director Julius Onah, who’s most famous for directing Netflix’s The Cloverfield Paradox and the critically acclaimed film, Luce. Though Joe and Anthony Russo directed the last two Captain America films, like Steve Rogers himself, they will step aside and pass the mantle of director to someone else. THE CLOVERFIELD PARADOX | PREMIERES TONIGHT | NETFLIX It was during this series that Torres inherited the title of Falcon from Wilson, along with his signature wingsuit, so audiences will undoubtedly see him take flight in New World Order. Likewise, Danny Ramirez will return to play Joaquín Torres, Wilson’s friend/ally in The Falcon and the Winter Soldier who had survived the chaos brought on by Thanos’s snap. But in the series’ final episode, Wilson had a memorial of Bradley built to honor his sacrifices for his country. government after he tried to stop them from eliminating many other Black soldiers secretly injected with the Serum to cover up their wrongdoings. However, Bradley was later imprisoned by the U.S. In this series, he was a Korean War veteran who was unknowingly injected with the Super Soldier Serum. For instance, Carl Lumbly will reprise his role from The Falcon and the Winter Soldieras Isaiah Bradley. This film will feature plenty of familiar faces. So everything that I do, I do as a common man.” Who is coming back to the MCU I just have wings on my suit that I got from the military. When discussing Wilson’s future in the MCU with Men’s Journal, Mackie compared his character to Chris Evans’s Steve Rogers, saying, “What you have to realize about Sam - he’s a superhero who is not really a superhero. Since Mackie’s character is leading the sequel, Marvel Studios seems set on presenting Captain America very differently in this film now that Wilson is the new Captain. But after Wilson fully embraced the title and the shield, his next adventure should follow him as he tries to make his country better as the new Sentinel of Liberty. The winged Avenger was unsure of himself becoming Captain America and representing a country that has horribly mistreated Black people for centuries. Anthony Mackie’s character took up the mantle of Captain America in The Falcon and the Winter Soldier, so this film will obviously be centered around Sam Wilson. Join method was passed a result from random.choices (which returns a list as result) and called on an empty string ( ' ' ) that acts as a separator. When calling random.choices we passed printable as the first argument and length (a parameter on our function) as the last argument. The first argument( population ) accepts an iterable and the last argument ( k ) specifies the number of items that will be randomly choosen from the given iterable. Random.choices takes couple of arguments but we are more concerned about the first and last argument. # generate random password and convert to string We will now create a python application to create a random password that meets the following constraints: The password is 12 characters long and contains at least one uppercase letter. It should combine random characters, numbers, and symbols. TypeError: 'str' object does not support item assignment A Python password generator is a simple program that can create random, strong, and secure passwords. If printable is passed to random.shuffle() without converting it first to a list, it'll result to a TypeError random.shuffle() takes a list as it's argument and changes the list item position randomly. Shuffling the list makes it difficult to have same password generated twice. Printable variable which holds an alphanumerical value (string: character sequence) is converted to a list (this is compulsory). # convert printable from string to list and shuffle Those constants are concatenated using python's F string and saved in a variable named printable. We already have some global variables declared, which holds string constants (digits, punctuation, characters(lowercase and uppercase) respectively). # create alphanumerical from string constants The above function is defined with one parameter length which defaults to 8 (the recommended minimum length of a password is 8). But…you can add a lot more to the feeding tube to make the process go faster. I didn’t use my machine on anything higher than stir and 2 I tried to go higher but the cheese starting flying a little much. You can use the bowl that comes with your kitchenaid but because of where the slices “come out” you’ll have to put the bowl off to the end to catch the slicings. LOL I don’t like a lot of prep work so the faster the better. To me taking down my kitchenaid off the shelf then getting the plug in blah blah blah- it’s just a lot. Otherwise the hand shredder or slicer can work for smaller dishes. I would suggest this product if you have a LOT of something to shred. The price point of this plug in isn’t bad (even for a frugal Franny like me). When I first grabbed that and my Kitchenaid mixer I was questioning whether or not it was really “that serious” but after I had shredded 5 blocks of cheese in a quarter of the time it usually takes- it definitely is. There are three: There is a larger one shredder, a medium shredder and then a large “slicer.” The functionality and ability to connect the blades to the Kitchenaid mixer is not overly complicated. What I like about the shredder from the outset is it has various blades. For the record I do have a hand shredder (because of course I do) but I wanted to compare it to the kitchenaid shredder. This past weekend I made my infamous SECRET mac n cheese (exclusive to those in the Peach Crew) and decided to use the slicer to slice the cheese. Canning can be a tedious project and because I’ve been slicing more and more cucumbers for more and more pickles (COMING SOON, lol) I needed something to help with that effort. I had been eyeing it for a while really for the slicing capabilities. This past Christmas my mother purchased the Kitchenaid Slicer/Prepper for me. The attachments weren’t that much more expensive but if I could do the same things for less money what difference did it make? Or so I thought. I had been wanting the Kitchenaid mixer for the longest but I just couldn’t click “purchase.” Of course my mixer came with the standard mixer heads but I hadn’t purchased any attachments (because frugal). My Kitchenaid mixer was a Christmas gift from Hubs (shout out to Him!) a few years (maybe 5 years) ago. So the combination of the those two traits usually makes me hesitate to buy things. But the surprise may come as where I am a kitchen product junkie- I am also kind of frugal. It should probably come as no surprise that I am a kitchen product junkie. Guitarron - Large bass guitar of Chile and Mexico. Directly related to medieval citterns and the English guitar. Closely associated with traditional Fado music. Traditionally played at feasts and weddings. Gaita - Spanish traditional bagpipe, usually with single drone, and plain leather bag. Has single drone, and 8 hole chanter, one hole for the thumb, 7 for the fingers. Gaida - Bulgarian version of mountain/shepherd pipes. Has three or four strings, very similar to a Rebec. Gadulka - Bulgarian type of fiddle played upright usually rests on knee. hence Tin Whistle, also known as penny whistle. Often made from a metal tube, either cylindrical or conical. Flat-backed instruments are commonly used in Irish, British, and Brazilian folk music.Fa'atete - Tahitian drum, covered in a tight, single membrane distinct, high sound like a drum roll.įipple - A word meaning the sharp edge of the lip (of a flute or recorder).įlageolet - Whistle, usually with 6 holes related to the recorder. Archtop instruments are common in American folk music and bluegrass music. Neapolitan mandolins feature prominently in European classical music and traditional music. Each style of instrument has its own sound quality and is associated with particular forms of music. The flat-backed mandolin uses thin sheets of wood for the body, braced on the inside for strength in a similar manner to a guitar. The archtop, also known as the carved-top mandolin has an arched top and a shallower, arched back both carved out of wood. The round-backed version has a deep bottom, constructed of strips of wood, glued together into a bowl. There are many styles of mandolin, but the three most common types are the Neapolitan or round-backed mandolin, the archtop mandolin and the flat-backed mandolin. Also like the violin, it is the soprano member of a family that includes the mandola, octave mandolin, mandocello and mandobass. The courses are typically tuned in a interval of perfect fifths, with the same tuning as a violin (G3, D4, A4, E5). It most commonly has four courses of doubled metal strings tuned in unison, thus giving a total of 8 strings, although five (10 strings) and six (12 strings) course versions also exist. A mandolin ( Italian: mandolino pronounced literally “small mandola“) is a stringed musical instrument in the lute family and is generally plucked with a plectrum. |
